T cell receptor signalling.

Upon engagement of the TCR by antigen presented on major histocompatibility complex (MHC) molecules, the Src family kinase Lck is activated and proceeds to phosphorylate immunoreceptor tyrosine-based activation motifs (ITAMs) on the ε, δ, γ and ζ subunits of the TCR. Phosphorylated ITAMs promote the recruitment and subsequent activation of another tyrosine kinase ZAP-70. Two known substrates of...

Stabilisation of β-Catenin Downstream of T Cell Receptor Signalling

BACKGROUND The role of TCF/β-catenin signalling in T cell development is well established, but important roles in mature T cells have only recently come to light. METHODOLOGY/PRINCIPAL FINDINGS Here we have investigated the signalling pathways that are involved in the regulation of β-catenin in primary human T cells. We demonstrate that β-catenin expression is upregulated rapidly after T cell...

Modelling the Tunability of Early T Cell Signalling Events

The Tunable Activation Threshold hypothesis of T Cells is investigated through computational modelling of T cell signalling pathways. Modelling techniques involving the π-calculus and the PRISM model checker are presented, and are applied to produce a stochastic model of T cell signalling. Initial results which demonstrate tuning of T cells are presented.

c-Cbl: a regulator of T cell receptor-mediated signalling.

The 120-kDa protein product of the c-Cbl proto-oncogene is a ubiquitously expressed cytoplasmic protein that is especially abundant in the thymus, indicating an important role for Cbl in thymic signalling. c-Cbl possesses a highly conserved N-terminal phosphotyrosine binding domain, a C3HC4 RING finger motif, multiple proline-rich motifs, and a number of potential tyrosine phosphorylation sites...

Notch signalling in the regulation of peripheral T-cell function.

The Notch signalling pathway plays a highly-conserved role in regulating the cellular differentiation and proliferation events that characterise pattern formation in the embryo. As cells in the embryo respond to environmental signals, similarly T-cells in the peripheral immune system must monitor their environment for antigens and respond accordingly by entering one of several potential differe...
